We're trying to track down a relationship shown up with my wife's (Annette D) Ancestry DNA match and would appreciate some help/advice.
Annette's DNA matches show Guy M as 1st cousin 1x removed or half 1st cousin (497 cM | 7% shared DNA), and his sister Daphne M as 1st cousin 1x removed (378 cM | 5% shared DNA). Paternal side
Guy & Daphne's mother was Margaret E (married to Willam M)
Margaret E's mother was Florence E, her father is unknown
It therefore seems likely, we think, that Margaret E's father was either Annette D's grandfather on her father's side, or one of his brothers.
I'm not well versed with cM & % - is it more likely to be Annette D's grandfather, or one of his brothers?

Three matches isn't enough to work with and answer the query I'm afraid so adding in shared matches is required. These might help point the way but much depends on the matches that are available.

Yes, I realise that more matches would help. We've found a distant cousin, descended from one of Annette D's grandfather's brothers, who's had a DNA test, just asking him if he'll share the results.

Think also in terms of the likely Most Recent Common Ancestor of your Wife and of Guy.
If you can build a tree around Guy going back to his Great Grandfather, expand it sideways as much as possible and then forward to the present day.
Look at shared matches and incorporate them into the tree as much as possible.
Try to ID other Cousins of Guy who likely share the same Grandfather, contact them and explain the situation.
Work various hypothesis to see the result.
I would start with the person who you think is your Wife’s Grandfather, in hypothesis 1 he would be the likely Father. See how the relationship fits.
I had a 240, 140 and 156 matches and it was enough to positively confirm relationships, contact with another person and them taking a DNA test finalised the relationship and shattered the brickwall.
It can be done.

Agree that the information is insufficient but other factors which may help are where Annette's grandfather and his brothers were at the relevant time.
I had a similar dilemma but the fact that 2 of the 3 possibilities had emigrated well before the relevant birth provided a clear solution to my problem

Unfortunately that doesn't help in this case, Annette's grandfather & his brothers were all living within about 8 miles of where Florence E was likely living, so it could potential have been any of them!

Using Ancestry Pro Tools I now have the shared DNA of Guy M and the great grandson of one of Alfred D's brothers, Gavin D, which is 275cM
Putting the cM figures into DNA Painter, which I've just discovered, gives a 71% probability that Margaret E's father was Alfred D
Does that seem reasonable?

That seems reasonable.
I would use it to build a tree and try to ID a series of people who could be approached with the suggestion that they take a DNA test.
It worked for me.

Well... I've now put in the shared DNA for Guy M and the daughter (Kaaren B) of one of Alfred's daughters (Pat D), the shared cM figure for her and Guy M is 230.
That changes the percentages to 48% probability that Alfred D is Margaret E's father, and 49% that Gavin D's great grandfather is Margaret E's father.
That seems to indicate that Margaret E's father was another of Alfred D's brothers I think? More DNA matched needed!

It's very difficult to distinguish from a set of brothers
I had a lucky break once when I learnt that 2 of the matches great uncles were half brothers and I could look for matches to the families of their respective mothers
2 were twins which also skews the results
Another time there was a big age difference between the single mother and the potential fathers . The younger of them was more likely especially as he was also a travelling salesman.
Distinguishing another set of brothers was by elimination as the grandson of 1 didn't have a high enough match to be a 1st cousin or half first cousin .
Good luck and keep us posted on your progress
I think this could be very useful practical example for others

Ps
If annettes parent or child tests you will get extra generation parameters to play with . This can also help eliminate some of the relationship possibilities
For example a child could either be a cousin 2R or a half C1R
Or a second cousin
Which may differ more widely in cm range
The parent could be a cousin .a half cousin or an aunt uncle to your match or even.a half sibling
Sorry if I'm stating the obvious
Or if my logic is flawed ...it's early morning here .

Just to tie this up..
I got a copy of Margaret E's birth certificate, no father's name, as expected. However it did have her address which turned out to be a Salvation Army hostel, the Salvation Army were able to provide me with the details of her time in the hostel and the 'alleged father' is named. Turns out to be Gavin D's great grandfather!
