I don't think that I have anything concrete to add to what you already know, but I too am descended from Peter Polland and Agnes Barclay, through their daughter Agnes. Until recently Agnes Barclay has been a brick wall for me as she does not seem to appear on any of the usual records.
However, within the last week I have been in touch with another descendant of Richard's, who seems to have a similar story about Agnes Barclay to the lead that you appear to be following and I would be very interested to find out what you know about her origins.
I do not yet have the details of my contact's story, but when I do I will post it here.

Here is what I have been given about Agnes by our cousin, via a friend of hers as she is not online herself.
Agnes was known as "Lady Agnes Barclay", living in Ireland. In her teens she met Peter Polland who worked on the estate, and they ran away together. When Peter and Agnes eloped, her father sent out a small army of men to search for them, without success. She dropped the "Lady" from her name, but after a while she missed the comforts she was used to and was no longer content with Peter whom she treated more as a servant than a husband.
My contact speculates that this is why she had left the scene by the time of the 1841 census.
Our cousin from whom this story came is, like you, a descendent of Richard Polland and Margaret Devine, through their daughter Margaret.
If this is basically the same as the story you have, it has obviously been in the family for a long time. Like you though, I haven't been able to find anything about Agnes from the limited searching I have done, but if it is true that she was titled I would have thought that the information ought to be there somewhere.
Hmm - there's nothing like a good mystery.
