I'm trying to locate Elizabeth Cook in Whitfield, Gloucester.
She was born around 1850 and was married to Henry Chapple in 1877 and living in Bristol by the 1881 census.

I can't find her in the 1851 census.
You wrote that she was born around 1850, so it's possible that she was born after 30th of march 1851 (the date the 1851 census was taken). I don't have acces to the 1861 :'( ,but someone else have, I'm sure.

The 1881 and 1901 census details suggest she was born in 1850. Hopefully the 1861 census will be helpful.

I don't seem to be able to find Whitfield in the registration districts for Gloucestershire. Can you give me an idea where it is? Maybe I'm just having an off day!

I found them on the 1851 at Thornbury -
William 1847, Mary 1817, Jane 1845, Elizabeth 1849, Charles 1812 and - not on the 1861 - Henry, big blot where age is but looks like a teenager (12?). He is blind.
